Practical Applications and Clinical Utility of PYLARIFY® (piflufolastat F 18) Injection: An ¹⁸F-PSMA PET/CT Imaging Agent for Prostate Cancer
A series of interviews was recently conducted with a panel of experts on prostate cancer imaging modalities that included Steven Rowe, MD, PhD, Associate Professor of Radiology and Radiological Science, Johns Hopkins University School of Medicine, Baltimore, MD, and Neal D. Shore, MD, FACS, CMO, Surgical Oncology/Urology Genesis Care, US; Director, CPI, Carolina Urologic Research Center, Myrtle Beach, SC. Read Article ›

The addition of 177Lu-PSMA-617, a radionuclide therapy that targets prostate-specific membrane antigen (PSMA), to standard-of-care treatment resulted in a 38% reduction in the risk for death versus standard of care alone in men with progressive PSMA-positive metastatic castration-resistant prostate cancer (mCRPC), according to findings from the phase 3 VISION clinical trial, which were presented during a plenary session at the ASCO 2021 virtual annual meeting. Cabazitaxel (Jevtana) is approved as third-line therapy for patients with metastatic castration-resistant prostate cancer (CRPC) after treatment with docetaxel and an androgen receptor inhibitor. Read Article ›

PARP inhibition is moving on from breast and ovarian cancer to the treatment of patients with prostate cancer and BRCA1/2 mutation. Read Article ›

Early results from the phase 2 TheraP clinical trial show that the targeted radiation therapy 177Lutetium PSMA-617 (Lu-PSMA) significantly improved prostate-specific antigen (PSA) response compared with standard cabazitaxel in men with metastatic castration-resistant prostate cancer (CRPC) that progressed after docetaxel therapy. Oral relugolix given daily is superior to standard androgen-deprivation therapy (ADT) with leuprolide (Lupron) in men with advanced prostate cancer, according to the results of the phase 3 HERO study, which were reported at the ASCO 2020 virtual annual meeting and published online before the meeting (Shore ND, et al. N Engl J Med. Read Article ›

Positron emission tomography (PET) scanning targeted to detect prostate-specific membrane antigen (PSMA) is poised to overtake conventional imaging, according to presentations at the 2020 Genitourinary Cancers Symposium. Read Article ›

Treatment with a combination of pembrolizumab (Keytruda) and enzalutamide (Xtandi) continues to show substantial activity in patients with metastatic castration-resistant prostate cancer (CRPC) who have already been treated with abiraterone (Zytiga), according to results of a recent clinical trial, which were presented during the virtual 2020 American Urological Association Annual Meeting. Circulating tumor DNA (ctDNA) analysis is a useful sequencing platform in metastatic castration-resistant prostate cancer (CRPC), and ctDNA alterations are detected in most patients with this type of prostate cancer, according to Justin Shaya, MD, Hematology/Oncology Fellow, UC San Diego Moores Cancer Center, CA. Enzalutamide (Xtandi) and apalutamide (Erleada) had strong showings in 2 separate, randomized phase 3 clinical trials demonstrating that these drugs delay disease progression when added to background androgen-deprivation therapy (ADT) in patients with metastatic, hormone-sensitive prostate cancer. Darolutamide, an investigational androgen receptor inhibitor, significantly improved metastasis-free survival in men with high-risk nonmetastatic castration-resistant prostate cancer (CRPC) compared with placebo in a large phase 3 clinical trial. Read Article ›

For patients with newly diagnosed metastatic prostate cancer, current standard management is androgen-deprivation therapy plus docetaxel (Taxotere). Contrary to previous assumptions, radiation to the primary tumor improves survival in men with low metastatic disease burden. This new finding came from the preplanned analysis of the multiarm, multistage STAMPEDE study presented at the ESMO 2018 Congress by lead investigator Chris C. Parker, MD, FRCR, MRCP, Consultant Clinical Oncologist, Royal Marsden NHS Foundation Trust, Sutton, England. The study was simultaneously published online in the Lancet.1 Read Article ›
